Efisiensi Penggunaan Enzim Taq Polymerase pada Pengujian Polymerase Chain Reaction

Polymerase Chain Reaction (PCR) merupakan uji diagnostik yang sangat sensitif. Enzim Taq salah satu komponen paling penting dalam pengujian (PCR). taq polymerase harganya mahal. Penggunaan pada PCR kurang efisien. Untuk mengetahui efisiensi penggunaan enzim dan volume terkecil berapa masih dapat digunakan metode PCR, maka dilakukanlah penelitian dengan membuat lebih kecil (5µl, 10 µl, 25 µl) dari standard (50 µl). Tiga isolat swab hidung anus anjing terinfeksi Parvovirus diekstraksi menggunakan Kit DNA Purifikasi. Hasil ekstraksi dijadikan Template PCR. Amplifikasi mesin Apply Biosistem Thermal Cycler. Visualisasi produk amplifikasi elektroforesis gel agarose 1%. menunjukan bahwa mampu memvisualisasikan baik. Pada 5µl, tidak semua sampel teramplifikasi Sampel 1 3 menunjukkan band tebal posisi sama kontrol positif. Sedangkan 2 adanya band. (10µl, µl 50µl) 3, gen memiliki panjang nukleotida 910bp (sesuai oligonukleotida), sedangkan juga tetapi posisinya rendah tipis 1, Adanya perbedaan tipisnya disebabkan oleh konsentrasi hasil ekstraksi. Perbedaan tinggi 2, berarti bukan spesifik parvo. Simpulan ini adalah virus parvo mengefisienkan 10µl direkomendasikan sebagai protokol untuk Virus Parvo.
